    The present invention relates to a composition comprising an autophagy enhancement compound. Small molecules that are able to enhance autophagy and lysosome biogenesis by activating the gene TFEB which can prevent the accumulation of toxic protein aggregates in treating neurodegenerative diseases are disclosed.     The Centers for Disease Control and Prevention recommend against treatment of malaria with chloroquine alone due to more effective combinations. In areas where resistance is present, other antimalarials, such as mefloquine or atovaquone, may be used instead.

Phosphorylation of TFEB at S211 enhances the binding of TFEB to 14‐3‐3 protein, thus favoring the retention of TFEB in the cytoplasm Martina et al, 2012. The lysosome regulates gene expression via TFEB. A Chloroquine treatment inhibits mTORC1 activity in primary hepatocytes. Primary hepatocytes isolated from 2-month-old Tcfebflox/flox control and Tcfebflox/flox;Alb-CreTcfeb−/− mice. However, at present, the functional changes of lysosome in the course of autophagy remain largely unknown. In this study, we provide evidence demonstrating the functional activation of lysosome achieved via a dual mechanism involving mTORC1 suppression and autophagosome-lysosome fusion.
